StepOne™ Software v2.1 supports both Applied Biosystems’ StepOne™ and StepOnePlus™ Real-Time PCR Systems. StepOne software allows the user to open and analyze experiments generated from either the StepOne or the StepOnePlus system. The software also enables the user to set up experiments, send experiments to the instrument, control the thermal cycling process in the instrument, collect data and analyze the collected data, all in an integrated and streamlined fashion.
· Experimental Design Wizards to help you design and set up experiments
· Pipetting protocols and recipes to set up experiments quickly
· Advanced set-up for expert users who require flexibility for more complex applications, such as multiplexing
· Quickstart set-up so you can begin a run immediately and enter plate information later
· Real-time monitoring of amplification growth curves enables you to view a run progress (can be viewed from a remote PC as well)
· Auto-baseline and auto-threshold for simplified data analysis
· Multiple Plots view for simultaneously assessing data from four perspectives
· Automated SNP genotype-calling with intuitive graphical output and quality-value assignment
· Troubleshooting flags to help you diagnose and solve problematic experiments
· Tool tips for easy identification of sample wells when viewing amplification curves or SNP genotyping plots
· Email notifications to alert you when a run has started or ended with the option to attach the completed run file
· Easy cut-and-paste functionality
· Export easily to PowerPoint®, Excel®, jpeg file, tab delimited text and PDF
· Perform runs with multiple temperatures on one plate using the VeriFlex™ Block thermal cycling mode (StepOnePlus only)
· Gene Expression Study Package to
– Analyze an unlimited number of plates in one study, only limited by the available memory
– View data by biological replicate group or technical replicate group
– Use multiple endogenous controls per study
– Enter and correct for known assay efficiencies
See accompanied End User License Agreement for details. You must agree to the terms of the license before installing or using the software.
· The software can open experiment files created with the StepOne software v1.0 , v2.0, v2.0.1 and v. 2.0.2
· When upgrading an instrument that is currently connected to a computer with StepOne software v1.0, a firmware upgrade will be required. The software will prompt you during the initial connection to perform the necessary upgrade and you will not be able to run the instrument until the firmware has been upgraded.
See Release Notes for details.
The computer hardware and operating system requirements for the StepOne™ Software are:
· Windows® XP with Service Pack 2 or later
· Pentium 4 or compatible, with minimum 512 MB of RAM (1GB for the gene expression study package) and 60 GB of hard drive capacity
· Minimum monitor resolution of 1280x1024
· One open Ethernet port for connecting to the instrument directly (co-located configuration)
· Internet Explorer 6.0 or higher (for online assay browsing and ordering)
· Microsoft PowerPointâ software (for direct export into PowerPoint slides)
· Logon to the Windows system with Administrator privilege
· Obtain the software installation package.
· Place the new version disk in the CD drive and allow the installer to launch automatically. If the installer does not launch automatically, double click the setup.exe file on the CD to start the installation.
· Agree to the End User License Agreement (EULA) when prompted and complete the installation
· The software will be installed, by default, at “D:\Applied Biosystems\StepOne Software v2.1” (referred to as the home directory of the StepOne™ Software)
· A program group, “Applied Biosystems/StepOne Software” will be created during installation. In addition, a short cut to the application launcher, “StepOne v2.1”, will be installed on the computer desktop.
· When a previous version of the software is detected on the system, the installer will perform an upgrade. The user will be prompted to proceed with the upgrade or to cancel the operation. To prevent data loss, it is strongly advised that all user data is backed up before upgrading the software.
IMPORTANT: To prevent data loss, it is strongly advised that all user data is backed up before uninstalling the StepOne™ software.
· From Windows “Start” button, find the “Applied Biosystems/StepOne Software” program group.
· Click on “Uninstall StepOne Software v2.1”.
· A wizard shows up. Follow the instructions and complete the uninstall operation.
· Optionally, rename or delete the home directory of the StepOne Software application. This ensures a clean environment for the next (clean) installation.
